چکیده

Rat small eye (rSey) involves a mutation in Pax-6 gene. The midbrain crest cells of rSey homozygote fail to migrate normally towards the frontonasal region resulting in absence of eyes and nose. In the present study, comparison of the distribution patterns of fibronectin, laminin, tenascin, chondroitin sulfate proteoglycan and HNK-1 epitope in craniofacial region between that of rSey homozygous and the wild type embryos at the day 11 of gestation were investigated. The results showed alteration of distribution pattern of HNK-1 epitope at the mesenchyme adjacent to the frontonasal ectoderm where the Pax-6 gene was strongly expressed. However, the other molecules did not show any obviously different distribution.

Codistribution of heparan sulfate proteoglycan, laminin, and fibronectin in the extracellular matrix of normal rat kidney cells and their coordinate absence in transformed cells

We used antibodies raised against both a heparan sulfate proteoglycan purified from a mouse sarcoma and a chondroitin sulfate proteoglycan purified from a rat yolk sac carcinoma to study the appearance and distribution of proteoglycans in cultured cells. Normal rat kidney cells displayed a fibrillar network of immunoreactive material at the cell surface when stained with antibodies to heparan s...

Ischemic injury leads to extracellular matrix alterations in retina and optic nerve

Retinal ischemia occurs in a variety of eye diseases. Restrained blood flow induces retinal damage, which leads to progressive optic nerve degeneration and vision loss. Previous studies indicate that extracellular matrix (ECM) constituents play an important role in complex tissues, such as retina and optic nerve. They have great impact on de- and regeneration processes and represent major candi...

Neuronal proteoglycans: biosynthesis and functional interaction with neurons in vitro.

Glycosaminoglycans (GAGs) with electrophoretic mobilities on cellulose acetate similar to heparin (H), heparan sulfate (HS), and chondroitin sulfate (CS) were detected in cell extracts and in conditioned medium of high-density, neuron-enriched cultures labeled with 35SO4. Heparitinase digestion revealed that heparan sulfate proteoglycans (HSPGs) were heterogeneous in charge density and were res...

Molecular forms, binding functions, and developmental expression patterns of cytotactin and cytotactin-binding proteoglycan, an interactive pair of extracellular matrix molecules

Cytotactin is an extracellular matrix protein that is found in a restricted distribution and is related to developmental patterning at a number of neural and non-neural sites. It has been shown to bind specifically to other extracellular matrix components including a chondroitin sulfate proteoglycan (cytotactin-binding [CTB] proteoglycan) and fibronectin. Cell binding experiments have revealed ...

1/31 Integrin on Neural Crest Cells Recognizes Some Laminin Substrata in a Ca2+-independent Manner

Neural crest cells migrate along pathways containing laminin and other extracellular matrix molecules. In the present study, we functionally and biochemically identify an o~1/~1 integrin heterodimer which bears the HNK-1 epitope on neural crest cells. Using a quantitative cell adhesion assay, we find that this heterodimer mediates attachment to laminin substrata prepared in the presence of Ca 2...
